I wanted to download Genshin Impact to my laptop but it says that I don't have enough space. I need the rest of the storage space for other things so I wanted to use a USB stick instead (I thought of 32GB because the game requires 25.5GB). Would it be possible to actually play the game?

Yes, you can, but it's not a nice solution. Better to upgrade the internal plate if possible.

You should always connect the stick to the same port. This increases the likelihood that the drive letter will not change. You have to adjust it if that happens.

Rather, free up memory where you would normally save something like this.
