My laptop has an 80Gb hard drive and a 380Gb hard drive. Windows is on the 80Gb hard drive. My question is whether I can install programs on the larger hard drive as well.

Try to move the memory swap file to the larger disk.
Are you sure these are not partitions? It doesn't really make much sense anymore to separate them. 80GB would be quite small for a hard drive
