As soon as the message comes that you should restart the PC
As a rule, Windows should have copied all the necessary data from the USB stick to the hard disk after the first message "Restart computer". For security, you can leave it in but also until the normal desktop comes for the first time.
After Windows has been installed, you can of course format the USB stick and use it elsewhere
