When installing Fortnite, you can choose the installation path. Then select a path on the USB stick. When the installation is complete, you can start Fortnite.
It is only important that you always connect the USB stick to play before you open the Epic Games Launcher. Otherwise the launcher can't find the installed game on the stick.

All drives or partitions (whether physical or virtual) are mounted under "/ Volumes" under their name. The absolute file path of a file e.g. A USB stick always stays the same, even if you plug it into another Mac. Even if you plug in several drives - as long as they don't have the same name.
